You can only unlock this quest in Evernight (Night) mode on Dainichi Mikoshi in Enkanomiya, as the quest giver will only appear in that setting.
Enkanomiya Day-Night Cycle Guide
You can start the quest by talking to the ghost underneath Dainichi Mikoshi. The ghost will have an unknown name, ''???'' and can be seen with another ghostly figure.
To get to the quest giver's location, go underneath an opening on the edge of the cliff by gliding southwest of Dainichi Mikoshi.

To look for Antei, you can easily get to the location by using the Phase Gate below the cliff of the Waypoint south of The Serpent's Heart! You can then walk and glide to the island where Antei is.
