Descending Moon unlocks automatically after completing The First Sliver of Moonlight Archon Quest, which is the first part of the True Moon Archon Questline.

Descending Moon Walkthrough
Talk to Marionette
Find Sandrone in front of the Curatorium of Secrets and talk to her about the formula she's been working on that will bring Columbina back. A complex meter will appear on the upper left corner of your screen that will progress as you move on with the Archon Quest.
If you open your map, you'll also notice several of Dottore's energy fields across Nod-Krai, inluding the newly added islands to the north—Voidsea Outlook, Wavechaser Plain, and Ashveil Peak.
Talk to Your Companions
Jump down to the plaza and speak with your companions Albedo, Durin, Flins, Nefer, and Lauma. After completing the three objective markers, Dottore will launch an attack on Nasha Town with his monsters.
Defend Nasha Town
The group will split into three pairs, so make use of the available elemental reactions to defeat the enemies. Since it will be raining during the fights, Hydro-related reactions are always enabled.
The Hydro Traveler, Lauma, Nefer, and Flins will fight the final wave of monsters for this objective. Aside from the healing orbs scattered in the plaza, Lauma's C1 effect can also provide healing for the party!

Defeat The Doctor
The Doctor will appear in front of the group after the last wave is taken out, and the fight against the new Weekly Boss will begin. This story-related boss fight will have a few gimmicks that will not happen in the Trounce Domain fight.
One such gimmicks is where The Doctor traps your allies one-by-one until only the Traveler remains. The captured character must survive in the space they're imprisoned in until it switches back to the Traveler to continue the fight.
The second phase of the boss fight, where Columbina sends you to another space, is also only available in the story. You'll only need to spam the Elemental Burst action whenever it is recharged to deal massive damage to The Doctor.
Finish off The Doctor with the help of all your allies through a series of quick-time events to complete the Archon Quest.

How Long Does Descending Moon Take to Finish

The Descending Moon Quest takes about 25 to 40 minutes to complete when speedrunning it, while it takes 40 to 50 minutes if you want to read every dialgoue in detail.
