The first thing you have to do is get to Enkanomiya. Once you're there, a quest chain will automatically be unlocked.
The Three Great Martial Trials can only be unlocked after completing the Entrance to Tokoyo, and the The Subterranean Trials of Drake and Serpent. Once you have the Bloodbranch Coral, go to The Serpent's Heart in Enkanomiya and look for a ''???'' ghost. You need to finish all the three other subquests under Erebos' Secret to access this quest!
You need to finish the other three subquests to unlock ''The Three Great Martial Trials.'' We have guides for each of the quests and their puzzles here:

The Three Great Martial Trials is the last quest in the Erebos' Secret quest line, this is where you get the rewards for completing the other three trials.
