You can use Event Hourglasses to replenish Event Stamina. Earn them by participating in events, specifically in beginner and intermediate solo battles.

You can also use your Poke Gold to replenish your Event Stamina. This option will be available by choosing ’Custom-select’ in the Event Stamina screen.
If you're out of Event Stamina, you'll need to wait for 12 hours to replenish a piece of your stamina.
This means that you need to wait for half a day before you can get one match against the AI if you don't want to use any Event Hourglasses or Poke Gold.
All New and Upcoming Events Schedule

You can use Event Stamina to participate in limited-time events to earn exclusive cards. Events such as the Lapras ex Drop Event require Event Stamina in the solo battles, but they will reward you with Promo-A card packs for each win.
Do note that losing or conceding during the event battles does not consume stamina as it is only used up whenever you win a match.
This means that if you lose against your AI opponent, you can always try again until you win!
