Complete Side Mission 22: A Call from Mable

You can obtain a Squirtle by completing the Side Mission, "A Call from Mable". This is the only way to get this Pokemon before unlocking their corresponding Wild Zone.

You can only choose one between Bulbasaur, Charmander, and Squirtle.

Squirtle Best Nature for Story

Bold or Calm

Due to Squirtle having very high Defensive stats, its best natures are either Bold or Calm to boost its Defense or Special Defense further.

Squirtle also has a better Special Attack stat compared to its Attack, so choosing one of the above natures allows Squirtle to still deal some damage with its Special Moves.

You can also give Squirtle either Relaxed or Sassy nature, allowing you to increase your Defense or Special Defense at the cost of reduced Speed.
