Since Kakuna is a Fast Physical Attacker, its best Nature is either Adamant or Jolly to boost its Attack or Speed further, while reducing its Special Attack stat.

You can also give Kakuna either Lonely or Naughty nature, if you will mainly use it for Story Content.
