These two bosses hold the key item needed to start one of the two ascencion trials, Trial of Sekhemas from Balbala and Trials of Chaos from Xyclucian. You'll be able to find Balbala in the Traitor's Passage and Xyclucian in the Chimeral Wetlands.

Players can unlock Ascendancy by progressing to Act 2 and 3 of the game. During these times, you'll be able to take on either the Trial of the Sekhemas or Trial of Chaos as part of the Ascension Trials.
Ascension is not missable as you can obtain Djinn Barya and Inscribed Ultimatum as random loot to go back and retry the trials should you fail your first attempt. Using higher level versions of these items to start a trial will allow you to get more Ascendancy points later on.

After defeating the boss of your chosen Trial, you can claim your rewards as well as the Ascendancy class of your choosing.
List of Classes and Ascendancies
The Trial of Sekhemas offers a Roguelike-style challenge. Your total EHP (effective health pool) will be used as your total Honour and everytime you take damage, your Honour will take a hit. This means that you'll need to take as few amount of damage as possible to complete the trial.

Upon reaching the Vastiri Plains, you can take on the Trial of Sekhemas where you will have to defeat Balbala, the Traitor.
If you enjoy Roguelikes that offer diverse challenges in exchange for valuable rewards, this Trial is ideal for you.

As you venture through the treescapes of the Vaal Jungle, you will come across the Temple of Chaos where the Trial of Chaos takes place.
If you prefer to do dungeons with dynamic challenges that feature mechanics that constantly change, this is the Trial to go for.

After the 0.2.0f patch update, you'll now be able to respec and even change your Ascendancy. To do so, you'll need to remove all of your Ascendancy skill points then clear a trial that would otherwise, award you with the same amount of ascendancy point you would have otherwise gotten from completing the trial.
