How to Unlock Duel of Destiny
Progress Shadow of Glory
You can unlock Duel of Destiny by progressing the main quest, Shadow of Glory, as this gives you access to the Septimont region.
The quest mark automatically appears on your map the moment you walk near it, however, you'll have to talk to Sulpicia first to trigger and start the quest!

Talk to Sulpicia
Duel of Destiny is located on Capitaline Hilltop, nearby the cable car system in city. Talk to Sulpicia who's standing a couple of steps from the edge, pondering about the supposed legendary warriors atop the city.
Find Artemus

At her behest, find Artemus on top of the city walls lining around the whole of Capitaline Hall. You can easily access this location by teleporting thru the Capitoline Hilltop Resonance Beacon, then walking all the way the elevator, and gliding towards the Artemus' location.
After you find Artemus, conduct the interview with him, then head next to his archnemesis.
Find Kaimastos
Kaimastos is located just across Artemus, on the same platform as him. They're quite far from each other, however, simply run towards the direction of Artemus until you spot Kaimastos. Then, conduct the interview as usual.
Report Back to Sulpicia
Once everything is in order, report back to Sulpicia on her original location. Exhaust all the dialogue options with her to conclude the quest!
