Regal Ancestor Spirit can be found after touching the Ancestral Remain in Hallowhorn Grounds (North). You'll need to first light six towers in the area to gain access to this Boss. The Ancestral Woods will be the nearest Site of Grace.

It will drop the Remembrance of the Regal Ancestor.

Charged heavy damage spells will be very effective

Spells such as Loretta's Greatbow and Comet Azur will be incredibly useful for this fight due to the Regal Ancestor Spirit's long attack animations as well as its Teleport ability. The latter ability presents a great opportunity to charge up these spells and deal immense damage since it'll take some time to reach you again.

Spam spells when the Regal Ancestor Spirit starts healing

Once the Spirit uses its Heal ability, back away then start spamming spells to counteract the gains to its health. While you won't be able to remove its life gains entirely, you'll hopefully be able to take out a large chunk of it. Night Maiden's Mist will be particularly effective since the Spirit will remain stationary, so the mist can deal max damage.

For melee users, the best place to attack the Spirit is underneath it and near its hind legs. You'll be able to avoid all of its antler attacks from here. Be careful when getting behind it since it can do a back kick that can knock you down.

Stay aggressive and attack constantly

You'll want to end the fight quickly by constantly attacking the Regal Ancestor Spirit to minimize the number of times it can heal. If you let the fight go on for too long, it can just heal enough times to outlast you.

Exploit its slow moveset and large size by doing several heavy attacks that deal immense damage. You can also coat your weapon in Magic Grease to increase your damage.
