Regal Ancestor Spirit can be found after touching the Ancestral Remain in Hallowhorn Grounds (North). You'll need to first light six towers in the area to gain access to this Boss. The Ancestral Woods will be the nearest Site of Grace.

It will drop the Remembrance of the Regal Ancestor.

Tips for Beating Regal Ancestor Spirit
Bleed attacks and weapons won't be that effective in this fight due to the Regal Ancestor Spirit's resistance to it
Try not to summon a Spirit Ash since the Regal Ancestor Spirit can also siphon health from these during its Heal ability
Use a shield that blocks 100% of physical attacks as well as the Barricade Shield ability to retain enough stamina to constantly attack the Spirit
Try focusing your attacks on its head to deal increased damage and to potentially stagger it

Charged heavy damage spells will be very effective

Spells such as Loretta's Greatbow and Comet Azur will be incredibly useful for this fight due to the Regal Ancestor Spirit's long attack animations as well as its Teleport ability. The latter ability presents a great opportunity to charge up these spells and deal immense damage since it'll take some time to reach you again.

Spam spells when the Regal Ancestor Spirit starts healing

Once the Spirit uses its Heal ability, back away then start spamming spells to counteract the gains to its health. While you won't be able to remove its life gains entirely, you'll hopefully be able to take out a large chunk of it. Night Maiden's Mist will be particularly effective since the Spirit will remain stationary, so the mist can deal max damage.

For melee users, the best place to attack the Spirit is underneath it and near its hind legs. You'll be able to avoid all of its antler attacks from here. Be careful when getting behind it since it can do a back kick that can knock you down.

Stay aggressive and attack constantly

You'll want to end the fight quickly by constantly attacking the Regal Ancestor Spirit to minimize the number of times it can heal. If you let the fight go on for too long, it can just heal enough times to outlast you.

Exploit its slow moveset and large size by doing several heavy attacks that deal immense damage. You can also coat your weapon in Magic Grease to increase your damage.

Melee Attacks

Melee Attack Variations
Antler Uppercut: Regal Ancestor Spirit starts glowing starts glowing and charges up an uppercut. You'll have plenty of time to avoid this since it'll bow its head down first.
Jumping Hoof Stomp: Regal Ancestor Spirit leaps up and jumps twice mid-air before slamming itself into the ground. Roll to the side to avoid this. Don't roll immediately after its second mid-air jump since it'll briefly float before landing.
Hoof Stomp: Regal Ancestor Spirit stands on its hind legs then stomps the ground. You can avoid this attack by rolling. An easy way to know when to dodge this is by watching out for when it puts down one of its back legs.
Glowing Charge : Regal Ancestor Spirit starts glowing then charges at your from a distance. Roll into the beast as it gets closer to you.
Antler Sweep: Regal Ancestor Spirit sweeps its antlers from side to side then shoves them at you. Roll against each attack.

Ethereal Flame: Regal Ancestor Spirit spits spirit flames at you. It can do this while jumping in mid-air or while on its feet. This has a limited range so you can just back away.

Heal: Regal Ancestor Spirit siphons health from Spirit Creatures in the area. This also deals AoE damage so keep your distance. Ranged users can attack it from afar to reduce the healing effect.

Teleport

Teleport Information
Teleport: Regal Ancestor Spirit falls to the ground and starts fading before teleporting away from you. While this can't be stopped, you can briefly attack the area it was previously at before teleporting and still deal damage to it.
