You can enter Ainsel River from the Ainsel River Well in east Liurnia. Exit the lift, then head southeast till you reach Uhl Palace Ruins. Then head northwest till you can see a large Malformed Star enemy that hurls gravitational magic at you. Enter the hall right under the Malformed Star by running and hiding behind obstacles. The map fragment for Ainsel River will be on a corpse near a Hermit Merchant.
Lake of Rot

Entering Lake of Rot rquires that you progress through Ranni the Witch's questline. After giving her the Fingerslaying Blade, the barrier blocking off the entrance to Renna's Rise will disappear. The waygate at the top of Renna's Rise will teleport you to a region of Ainsel River that leads to Lake of Rot.
From here, you must head all the way to the west of Nokstella, Eternal City, where there will be a lift that takes you down. Follow the path a little bit more going south, and you will finally reach the Lake of Rot. The map fragment will be on the shore, close to the first Site of Grace you see.

Ainsel River Scarabs
Teardrop Scarabs are scurrying little creatures that carry rare and useful items and skills. Crimson and Cerulean Scarabs restore either your HP or FP. Make sure to defeat them before they run away and disappear!

Ainsel River Sacred Tears and Golden Seeds

Sacred Tears increase the potency of your Flasks, while Golden Seeds increase your Flask's number of uses. These are two very useful bolstering items that will help keep you alive longer.

A Golden Seed can be found under an Erdtree sprout up on a cliff. To get here, you must traverse the upper ramparts of Nokstella till you reach the topmost building on the west side. Then find an elevator to take you down to the Golden Seed.
Ulcerated Tree Spirit in Lake of Rot

An Ulcerated Tree Spirit mini-boss will drop a Golden Seed if you kill it. This mini-boss can be found in Grand Cloister in the Lake of Rot.
