Farming Duriel, Maggot King and Andariel, Maiden of Anguish is one of the most reliable way to obtain the Rod of Kepeleke as both of these Uber Bosses include this Unique Quarterstaff in their Drop Tables. Keep in mind that Lair Bosses no longer drop loot on death but instead requires you to open the hoard chest using Lair Boss Keys after defeating them.
Therefore, each time the hoard drops a guaranteed Unique Item, there is roughly a 1/8 or ~13% chance that it will be the Rod of Kepeleke. To increase your chances, defeat these bosses and open their hoards on higher Torment difficulties for more Unique drops.
Lair Boss Loot Tables (Season 8)
As of Season 8, summoning materials or Lair Boss Keys are no longer shared throughout the party as you need to spend your own materials in order to claim the hoard chest after defeating the Lair Boss. You can use the Party Finder to look for players that you can trade Lair Boss Keys with or help you farm Echo of Duriel, Echo of Andariel, and other Lair Bosses more efficiently, speeding up your hunt for the Rod of Kepeleke.
How to Use the Party Finder Feature

The next best method to farm for the Rod of Kepeleke is by trading Murmuring Obols for Quarterstaffs at the Purveyor of Curiosity. While the Quarterstaff you receive will have random quality, it's not uncommon to get a Unique Item from this method. Since there are only two Unique Quarterstaffs in the game, it is likely you'll get the Rod of Kepeleke through this method compared to other Uniques.
Clearing higher tiers of the Pit of the Artificer allows you to quickly farm large amounts of Murmuring Obols. This is an excellent way to gather Obols to trade for Quarterstaffs and possibly get the Rod of Kepeleke.

In the Kurast Undercity, you can use the Unique Tribute which rewards multiple Unique Items upon successfully clearing Kurast Undercity. While the Unique rewards are random, this is a fairly reliable method for farming the Rod of Kepeleke.
