Glyphs are primarily obtained by defeating enemies in Nightmare Dungeons. The higher tiers you tackle, the higher the drop chances of Glyphs so we highly recommend trying to get all your Glyphs once you are able to clear Nightmare Dungeons easily.
Another method of obtaining Glyphs are by completing objectives and getting your cache rewards from the Tree of Whispers. The Tree of Whispers Cache can drop glyphs occassionally which can give you a headstart when collecting glyphs for your current class.
How to Complete the Tree of Whispers
Glyphs can only drop while playing on World Tier 3 or higher. Even if you clear Tree of Whispers during World Tier 1 or 2, it will be impossible to get Glyphs as rewards. Therefore, its highly recommend to power through the earlier world tiers and head straight to higher tiers if you wish to get Glyphs as soon as possible for your character.

Glyphs are items that you can socket in to empty Glyph nodes in your Paragon Boards. These Glyphs can each grant unique effects to your character and build depending on the glyph used as well as the existing nodes you unlock inside the Glyph's radius.
To further strengthen your Glyph effects, you'll have to meet a secondary requirement for each Rare Glyph to activate their secondary effect. Failing to complete the Glyph's requirement will only cause it to grant its first effect making the Glyph suboptimal for your build.
