This request is unlocked after receiving The Slumbering Lord of the Tundra mission in the main story. You must also have completed the previous request given by Pesselle, Request 34: Croagunk's Curative Poison, and talked to her afterwards about the results of the curative poison.
The Slumbering Lord of the Tundra Walkthrough
One of the unlock conditions for this request is to talk to Pesselle about the results of her previous request. To do this, you'll need to talk to her during the day in Galaxy Hall, since she goes to a different location at night, after completing Request 34.

Medicinal Leaks are incredibly common out in the Obsidian Fieldlands. However, since you will require 100 of them to complete the request, it is unlikely that you can collect this much in a single visit. Instead, it is best to collect the Medicinal Leaks as you continue through the story and do other tasks.
Instead of going out into the field to gather the Medicinal Leaks, you can buy them instead from Ginkgo Guild Merchants. Each leak will cost ₽120, so purchasing all 100 will cost a total of ₽12,000.
