You need to finish all four temples and all four Thyphlo Ruins Side Adventures first before you can unlock the Investigate the Thyphlo Ruins Side Adventure.

The Dusk Claymore can be found in a chest next to the monolith at (0385, 3169, 0164) coordinates inside the newly risen structure while progressing through the "Investigate the Thyphlo Ruins" side adventure.
Get it From Scanning the Ganondorf (Super Smash Bros.) amiibo
You can get the Dusk Claymore by scanning the Ganondorf (Super Smash Bros.) amiibo.

How to Scan amiibos
Scan an amiibo by making sure amiibo scanning is enabled in the Options Menu. Then, press and hold the L button, select the amiibo icon in the Tool Wheel, and tap your amiibo on the NFC Touchpoint of your controller.
Exchange 140 Poes to Bargainer Statues in the Depths
The Dusk Claymore will be available at any Bargainer Statues in the Depths for 140 Poes once you've received the weapon in the side adventure, "Investigate the Typhlo Ruins" or by scanning the amiibo.
You need to have interacted with at least one Bargainer Statue in the Depths for it to be available. The Dusk Claymore is unavailable for exchange at the Lookout Landing's statue.
