Harlow's Last Stand is unlocked as part of Natsai Onai's Relationship questline. In order to unlock it you'll first need to have completed all of the previous quests as listed above.
Additionally, although not technically part of the Natty questline, you'll also need to complete the main quest Wand Mastery.
Natty is at the quest starting location in between the Floo Flames of Bainburgh and West Manor Cape. She'll be found on the beach only at night. If you go there at any other time, Wait in the glowing circle to fast forward the time.
How to Skip Time: Day and Night Cycle Explained

You'll be transported to the Hospital Wing after the boss fight. After the quest ends, the next quest Acting on Instinct will immediately begin. Best to do that right away, since it completes the questline.
Harlow is the only enemy you need to defeat during the boss fight. There's no need to waste your time fighting the other Ashwinders.
