Mesprit appears in the cavern in the middle of Lake Verity. It only appears after defeating Team Galactic at Mt. Coronet and either catching or defeating Dialga or Palkia.
Unlike the other Lake Guardians, interacting with Mesprit doesn't start an encounter. Instead, it will exit the cavern and start roaming the Sinnoh Region. Interacting with Mesprit automatically adds it to the Pokedex.
The Marking Map app on the Poketch will show you which area Mesprit roamed to. This makes it easier to track it down even if it runs away from battle.
How to Get the Marking Map App
Mesprit will change locations everytime you enter a new area. Chasing Mesprit all over the map will give you a hard time as it might fly away to a far area once you enter a city/town or Route.
Instead of using Fly or riding your Bike to across the map to catch Mesprit, you can simply go to a city or town and run to the adjacent Route back and forth until Mesprit appears on the location. Once that happens, use Repel and enter the grass patch to start an encounter with Mesprit.
Floaroma Town and Route 205 are great locations to do this as there is no gate that separates the two areas.

Mesprit can be tricky to catch due to its nature of being a roaming legendary. When tracking down Mesprit with the Marking Map App, we recommend having a bulky Pokemon in the lead who can keep Mesprit in battle through the use of the move Block.

When facing Mesprit, we recommend using either Snorlax with Block and Yawn or Bronzong with Block and Hypnosis. These Pokemon give you a reliable way to prevent Mesprit from escaping, and access to a status-inflicting move to improve catch rates.
Using Block prevents Mesprit's escape as long as the user doesn't faint or switch out. Take note that you can use Block before Mesprit flees even if your Pokemon is slower.
Bronzong is the most accessible and best choice here thanks to its 4x resistance to Mesprit's Psychic-type moves, Extrasensory and Psychic. It also has access to Hypnosis which will let you put Mesprit to sleep for a better chance of catching it. Just be careful not to knock it out!
The Ultra Ball is a reliable go-to ball to use. If you are battling Mesprit at night, the Dusk Ball becomes the better choice.
If about a dozen turns have passed, you may also rely on the Timer Ball for higher success rates of capture.
As a last resort, you may also opt to use the Master Ball for a guaranteed catch without worrying about the hassle of locking Mesprit down.
Mesprit is also obtainable in Pokemon Legends: Arceus during the course of its story.

Mesprit, as well as the other Lake Spirits, are not shiny locked so make sure to save before engaging it in battle.
You can try catching a shiny Mesprit by soft resetting. It may take you several resets before a shiny shows up, so make sure you're ready with the right Pokemon and a healthy stock of Poke Balls in your bag.

Mesprit's balanced offensive and defensive stats and above average Speed make it a great fast support Pokemon, making Timid a good choice for Nature. Modest is also a good Nature if you want a more offensive-oriented Mesprit.
Note: While Mesprit also has a decent Attack stat, its limited physical movepool and lack of Attack-boosting move makes it a poor physical attacker.
