All items that are exclusively found in each of the three major areas (Village, Castle, and Island) are considered missable. The game has no chapter select feature, nor any way to return to the previous area, so ensure that you've collected everything needed before progressing to the next region.
However, weapons and other items sold by the Merchant are not missable, as you can still buy them from him in every shop found later in the game.
Clockwork Castellans are easy-to-miss collectibles scattered throughout the Resident Evil 4 Remake. There's always one Castellan that you can find per chapter, so keep an open eye and ear out for them as you play through the game.

Requests are optional side missions that you can find throughout a chapter. These include requests for the Blue Medallion collectibles.
You can only complete the objectives for each Request in the same chapter you found them, meaning they must be finished before moving on to the next chapter.
There are treasures scattered around every area of the game, usually found inside chests, drawers, and barrels. All of these valuable items are missable, as you cannot go back to them after progressing to the next region.

Leaving the Quarry after the El Gigante fight in Chapter 4 is a point of no return, as you will not be able to return to the Lake past this point.
Crossing the bridge leading toward the abandoned house near the end of Chapter 5 is a point of no return, as you cannot return to the Village past this point in the game.
Using the elevator after taking down the Verdugo in Chapter 10 is a point of no return, as you won't be able to return to the Sewers past this point in the game.
After reaching the Clock Tower, riding the lift near the top is a point of no return, as doing so will prevent you from backtracking through the Castle.
Opening the gate past the Merchant that starts the fight with Ramon Salazar is a point of no return, as finishing the boss fight will force you to ride the boat to the Island, preventing you from returning to the Castle.
Going past the Merchant at the start of the chapter is a point of no return, as the path leading forward leads to the Saddler boss fight, which continues toward the end of the game.
The Merchant will tell you when you’re about to cross a point of no return. When he says, "Might want to take care of any leftover errands before going this way. Be a shame to live the rest of your life wondering 'what if?'—am I right?", make sure that you've explored and looted everything beforehand before moving forward.
