You can get Destiny Knot in Route 224. Note that you can only access this route when the guy blocking the path in Victory Road leaves which happens when you obtain the National Pokedex.

How to Unlock the National Dex

Pokemon in your party with the Ability Pickup have a chance to obtain Destiny Knot after a battle.

Do remember that only Pokemon in your party has a chance of obtaining an item. Additionally, you have to make sure that the Pokemon has no held item for it to have a chance of picking up the item.

Effect An item to be held by a Pokemon. If the holder becomes infatuated, the opposing Pokemon will be, Too.

Breeding Item

When a Pokemon holding Destiny Knot lays an egg, five of the twelve IVs from both of its parents will be passed down to the child Pokemon.

Therefore, if both parent Pokemon has high IVs, their offspring will have high IVs as long as one Pokemon is holding Destiny Knot.

Make a Pokemon hold Destiny Knot when you leave it in the Pokemon Daycare.

How to Train and Breed Pokemon

The Effect of Destiny Knot Does Not Stack!

Even if you have two parent Pokemon both holding Destiny Knot, the effect will not overlap. The child Pokemon will still receive only five IVs from the parents. It is not possible to completely guarantee that their offspring will get you preferred IVs when both parents are holding the item.

Pokemon with the pick up ability can get the destiny knot!! I already have 2 and haven't got this one mentioned in the article!
