Messmer's Kindling is obtained by defeating Messmer the Impaler at the top of Shadow Keep.
To get to Messmer, reach the Specimen Storehouse in Shadow Keep and progress through the area until you reach the Dark Chamber Entrance Site of Grace.

Messmer's Kindling is used to burn the Sealing Tree located south of the Church of the Bud in the Ancient Ruins of Rauh. The path to the tree is only accessible after defeating Romina, Saint of the Bud in the church.
Burning the Sealing Tree will open up the entrance blocked by shadow roots in Belurat leading to Enir-Ilim. To get to this entrance, take the elevator east of the Stagefront Site fo Grace and head up the top of the stairway.
Note that burning the Sealing Tree will lock you out from completing NPC questlines in the DLC, so make sure to wrap up everything by then.
