To be able to use Rykard's Great Rune, which you get for defeating Rykard, Lord of Blasphemy, it first needs to be activated at the Divine Tower of Altus. Once you reach the top of the Divine Tower of West Altus, you'll be able to activate Rykard's Great Rune.
