Most of the side quests are unlocked after reaching level 60 and changing the difficulty to Hell I. From there, you can visit the Adventure Seeker in Westmarch to accept side quests.
You will be given three side quests daily for a specific zone that will refresh at 3am (server time). You can also pay the Adventure Seeker with gold to change the available quests. After you accept the side quest set from the Adventure Seeker, it won't disappear until you complete its objectives or abandon the quest.
In addition to the quest rewards, completing a side quest also spawns a Resplendent Chest containing pieces of equipment and some Enchanted Dust.
Clearing all the side quests of a zone is needed for its Exploration Achievement completion.
Achievements Guide: All Rewards and How to Unlock
There are also side quests that randomly pop up while players are exploring a zone, whether they're in the middle of a main quest, farming in the area, or just passing through. There are only four quests of this kind, and all of them involve killing enemies that vary based on the zone and area.
Completing one also rewards you with a Resplendent Chest containing 2 pieces of equipment and 3 Enchanted Dust.
For this random side quest, you will need to kill a certain number of enemies inside the area marked on the map. You must reach the required number of kills without leaving the area, as doing so will automatically fail the side quest.
A unique boss monster will spawn nearby that you will have to defeat to obtain a Resplendent Chest. The boss will be marked with a purple skull on your map and can also be seen and attacked by any player.
However, the reward for defeating the unique boss only goes to the player who triggered the side quest. If by chance you run into one of these Wanted monsters and you notice their health bar is colored gray, you won't receive anything once you take it down. It would still be nice to lend a hand, though!
Another thing to remember regarding Wanted side quests is, although they are still triggered by chance, their locations and corresponding unique boss will remain the same.
This side quest is activated by interacting with a Cursed Chest. Trying to open one of such chests will spawn spirits around you. You will have a bonus objective to defeat these enemies within a time limit, which will reward you with another Cursed Chest if done successfully. Failing to do so will still unlock the Cursed Chest, but without the extra chest.
A group of enemies will spawn around your current location. Defeat them to get your rewards.
