In the Cargo Hold of Shinra-8

Kaiser's Knuckles can be obtained during Chapter 5 of the main story. It will be found inside a chest during the segment where Cloud and Tifa explore the Cargo Hold of the Shinra-8. Climb a small set of stairs to find the chest.

The Kaiser Knuckles weapon can be bought from a store that sells armor starting from chapter 6 onwards.
