You can earn Mastery Points by completing quests, eliminating targets, and participating in activities in Assassin's Creed Shadows (AC Shadows). Read on to learn all the methods to obtain Mastery points.

Each Samurai Daisho you kill in a Castle grants one Mastery Point. Some Castles have more than one Samurai Daisho and clearing out Castles earns a lot of XP and rewards you with loot and Legendary equipment.
All Castle Locations and Rewards
Assassination targets reward you with 2 Mastery Points after you kill them. This is a good way to start accumulating Mastery Points especially in the early game when at least 2 objective boards become available - The Kabukimono and the Butterfly Collector.
You gain Mastery Points as you progress through the main quests and accomplish side quests. When you gain enough XP, a notification should appear at the center of your screen whenever you obtain one Mastery Point.

The simplest way to gain Mastery Points is to grind by accomplishing contracts, completing activities such as praying at Shrines, finding Lost Pages in Temples, or exploring Kofuns. You also gain XP from exploration.
