Near the end of the Arena questline, you will come face to face with Agronak gro-Malog, also known as the Gray Prince, reigning Grand Champion. If you defeat him, you will be locked out of the Origin of the Gray Prince questline.
Completing this quest will reward you with the permanent ability Gray Prince's Training, which grats a +3 to your Athletics, Blade, and Block. Additionally, completing this quest will make the final fight against the Gray Prince easier, as he will not fight back.

M'raaj-Dar, the Khajiit mage and merchant of the Dark Brotherhood, sells several unique items, including the Eye of Sithis, Hands of Midnight, Bow of Infernal Frost, and Perditions Wrath. He is also an easy source of Poisoned Apples.
If you want any of these items, make sure to buy them from him before you complete the Dark Brotherhood quest The Purification.
How to Join the Dark Brotherhood

Although you can become a vampire in numerous ways, the easiest way would be to receive the Dark Gift from Vicente Valtieri, who will offer it to you after you climb the ranks in the Dark Brotherhood.
Similar to M'raaj-Dar, this easy way to become a vampire will become unavailable once you complete the quest The Purification.

These are some side quests that you can miss or get locked out from completely once you reach a certain point in the game. Make sure to get these quests as they have some useful rewards.

The quest The Purification will have you purging all of the members of the Dark Brotherhood who reside in the Cheydinhal Sanctuary.
While you can get some good loot from the bodies, you will no longer be able to buy items from M'raaj-Dar or become a Vampire thanks to Vicente Valtieri. Make sure to do these things first before completing the quest.
The Kvatch Oblivion Gate is the first required gate for the main quest line. Saving Martin from inside the Gate and finishing the quest will have the game start randomly spawning Oblivion Gates throughout the map.
If you do not want to deal with the random Oblivion gates, you can choose to postpone doing this quest until you have done everything you wanted to do.
In the Main Story quest Blood of the Daedra, Martin will ask you to bring him any Daedric Artifact. It is important to note that whatever Daedric Artifact you give him will disappear for the rest of the game. Choose what artifact to give him wisely.
Light the Dragonfires is the final quest of Oblivion's main questline. Once you complete this quest, all open Oblivion Portals will automatically close. That means you will no longer be able to explore them to get their skill exp and loot. If you want to maximize your experience and loot, make sure to close all of the Oblivion Portals before starting and completing this quest.
During the Shivering Isles questline, in the quest The Cold Flame of Agnon, you will be asked to decide if you want to become the ruler of Mania or the ruler of Dementia. This will decide which achievement you get (Duke Dementia or Duke Mania), as well as what enchantments your Ring of Lordship will have.
If you want to get both achievements, save before doing the Ritual of Mania or Ritual of Dementiua quests, and reload to get the other one.
