Breaking Glowing Spot are the only way you can obtain Mysterious Slates. If the Glowing Spot is in front of you, you must use Rock Smash in order to destroy and break the block open.

Ancient Artifacts such as Lost Relics, CDs, Fossils, and other rare Materials can also be acquired by breaking Glowing Spots, so be prepared for a lot of tradeable and giftable items.

The Dowsing Machine is used to search for more Glowing Spots. In order to start searching, you must first have a Pokemon that has a Search Specialty to follow you.
We recommend the ones that can fly or float since the landlocked Pokemon can get stuck if you swim or fly around.
Dowsing Machine Guide: All Pokemon with Search Specialty

Giving Mosslax Bitter food, like a Salad with a Leaf and Seaweed as Ingredients, will increase the odds of finding rare items! Try to plan out your searches when feeling him to not waste time.

During Lucky Days, Glowing Spots are more frequent, giving you more rewards to reap from exploring during this time.

Glowing Spots will refresh everyday, so do make it your daily task to dig up as many per map. If you don't mind changing the date, you can increase your efficiency by continuously changing it to Sunday.
Time Traveling Guide: How to Change the Time of Day

Despite being able to take a photo of the blocks, you cannot 3D Print these blocks to cheat your way from looking for them.

Even after completing the Mural, Mysterious Slates will still spawn in your world. You can sell these via Trades for easy barters!

※ Hover over the corresponding Slate to see which Unown shape they have on their back!
You can set the block on the ground and move your camera to the back of it to see which one it is. Alternatively, if you have all of them in your inventory, you can also auto-sort them and they will get into the proper order.
If you want to make it difficult, you can also solve it as a regular puzzle. The block will have a short sparkle animation if you get the placement right.

You can attach Mysterious Slates that you find around the area to the Mysterious Mural. You will need to match them according to the corresponding Unown Alphabet found on the back of the slate.
Attaching all 27 Slates to the correct symbols will unlock the Pokemon, Mew, as a townsfolk.
How to Complete the Mysterious Mural
The Mysterious Mural can be found northeast of the Pokemon Center in the Withered Wasteland. You will need to break the wall there to find the hidden area.
Afterwards, break the wall to the left where you see the chiseled blocks. This will uncover a set of stairs going down, which leads to the Mysterious Mural.

Other than the mural, the Mysterious Slates will do nothing than just be bag filler. You can put them on the ground and check behind to see what letters they correspond to though.
Either stuff them in a Storage Box until they're completed, or you can come back to the Mural everytime you pick one up and slot them in there one-by-one.
