Moonrise Towers is located in the Shadow-Cursed Lands where much of Act II takes place and is a locale shrouded in life-eating darkness that makes it normally impassable without a light source or blessing.
Getting to the Shadow-Cursed Lands requires you to go through either the Underdark or the Mountain Pass.
Your path to the Shadow-Cursed Lands depends on your choices in Act I and the factions you've decided to side and help.
Siding with the Goblin Camp's Absolute Leaders will take you through the Mountain Pass and a direct path to the Shadow-Cursed Lands.
If you side with the Druid Grove, you'll need to head down the Underdark and through the Grymforge to eventually get to the Shadow-Cursed Lands.
Underdark or Mountain Pass Route?
If you side with the Absolute Leaders, you will immediately obtain a Moonlantern from the drider Kar'niss. Otherwise, helping the Harpers in the Last Light Inn will allow you to obtain the Moonlantern if you sided with the Druid Grove.
Regardless of your choice, it's better to free the pixie once you obtain the Moonlantern since the Pixie's Blessing she'll give allows you to traverse the Shadow-Cursed Lands freely without having to hold the Moonlantern!
You can reach Moonrise Towers by following the path southwest from the Last Light Inn and through the Tollhouse.
The Moonrise Tower Waypoint is located on a wall just before the wall of shadow obscuring the bridge to Moonrise Towers.
Once you arrive at Moonrise Towers, Adept Malik will hail you and identify you as a True Soul and will be presented by five dialogue options.
The first four replies will give you full access to Moonrise Towers to explore, while the fifth dialogue option Great News: I'm here to take your head will cause the Moonrise Towers npcs to turn hostile.
