Specializations are unlocked during the final main story mission, Secrets of the Vault. Defeat the 2nd boss during this mission to unlock the Specializations tab in your ECHO menu.

You can respec Specializations by spending 1,500 Eridium at any Respec Machine found in major towns.
It’s currently unknown whether Specialization has a level cap, though it’s likely very high if it does. We recommend spreading out and investing in other skill trees instead to make the most of different Specializations.
It’s best not to invest all your levels into a single tree anyway, since diminishing returns make each additional point less effective as you invest further.
