The weekly reset is every Thursday, 9:00 AM, PDT. Depending on your platform and version, it may take some time for the game to properly reset its weekly content.
You can access the two Weekly missions, the Wildcard Mission and Moxxi's Big Encore, directly from the Matchmaking menu.

A Wildcard Mission is a much more difficult replay of a main mission where every enemy has modifiers. After completing it, you will be rewarded with a Legendary item.

Every week, a Moxxi Encore Machine will offer a harder version of a boss. They will drop more loot, raising the chance of dropping their Legendaries. However, instead of cash, you have to pay Eridium.

The Black Market is a vending machine that trades cash for Legendary Weapons. Its location will change every weekly reset.

To unlock weekly content, you have to finish all the main missions. Afterwards, you have to complete a set of new missions that will unlock the weekly content and your first Ultimate Vault Hunter rank.
