Vex
Vex is, as of writing, the strongest character in the game, especially for single player. Two of her action skills, Phase Phamiliar or Dead Ringer, can handle large groups of enemies. Her third Action Skill, Incarnate, can be used to heal her and deal massive AoE damage.
Her Vault Hunter Trait will imbue her Action Skills with the element of her equipped weapon. This means she can exploit any elemental weaknesses should you have the right gun equipped.
Best Vex (Siren) Build and Leveling Guide
Rafa
Rafa is this game's brawler who focuses on gun and/or melee damage. His Action Skills will equip an additional weapon to ramp up your damage, like his best one: the Peacebreaker Cannons.
Since he prefers close-range combat, he needs good defensive gear and ways to sustain his shields and health to fully dominate the battlefield.

A Tier Vault Hunters
Amon
Amon is a frontline tank with high durability and close-range damage. You can play a defensive tank build with Scourge or focus on freezing and burning enemies with Crucible. He can take a lot of punishment and dish it back out.
However, his Cyro and Incendiary damage can be a limiting factor since it is very weak against shields. Always bring a Spark weapon to cover his weaknesses.

B Tier Vault Hunters
Harlowe
Harlowe is the support character of the cast. Her Action Skills like Flux Capacitor can debuff enemies while providing buffs. However, her Action Skills are best used when you are playing with a party. In single-player mode, she is outclassed by other characters.
Additionally, her Elemental Affinities, Cyro and Radiation, are not super effective against any type of health. She needs to use Maliwan weapons to take advantage of elemental weaknesses.
