The National Dex includes all Pokemon from the Kanto, Johto, and Hoenn regions. While some of them will appear in the game, many of them are locked to Pokemon Ruby, Sapphire, and Emerald.
It is important to note that this was impossible to do in the original FireRed and LeafGreen releases in the Game Boy Advance console, as some of these Pokemon cannot be transferred into the game.
To complete the National Dex, you must trade with players who have Pokemon Ruby, Sapphire, and Emerald, along with some GameCube games, namely Pokemon Colosseum and Pokemon XD: Gale of Darkness. However, these games are tied to older Nintendo devices and aren't compatible for trading with the Nintendo Switch.
While it is likely that Nintendo Switch ports of Pokemon Ruby, Sapphire, and Emerald may release, there is currently no way to trade with these versions.

In FireRed and LeafGreen, you can complete the regional Kanto Dex, which requires you to capture 150 Pokemon across both versions. This doesn't include Mew, as this Pokemon is only available from Emerald and is mainly an event-exclusive Pokemon.

The National Dex unlocks certain features that were previously locked. This is considered post-game, as the majority of them are tasks you can accomplish after beating the game.

The National Dex gives you access to some Pokemon from Gen 2 and Gen 3, with a majority of them seen in the Sevii Islands. Furthermore, some Pokemon Evolutions are tied to Gen 2 and only become available once the National Dex is unlocked.
Take note that there are still a lot of Pokemon in the National Dex that are missing in FireRed and LeafGreen. Furthermore, you will need to trade to get some of the version-exclusive ones!
Version Exclusives and Differences
Upon receiving the National Dex, it also gives you access to the rest of the Sevii Islands after giving Celio the Ruby and receiving the Rainbow Pass. This is an area previously inaccessible before becoming the Champion.
There are certain Non-Kanto region Pokemon you can encounter after receiving the National Dex. These Pokemon are mostly available in the Sevii Islands, which you can capture in the post-game.

To unlock the National Dex, you must become the Champion in the Kanto region. Defeat the Elite Four and the Champion by progressing through the story.
Elite Four and Champion Walkthrough Guide
One of the requirements to receive the National Dex is to have gone to the Sevii Islands. Visit One Island before returning to Professor Oak in Pallet Town.

Once you complete all the requirements and have registered at least 60 Pokemon in the Kanto Dex, return to Pallet Town and talk to Professor Oak. He will give you the National Dex, requesting that you discover the rest of the Pokemon available in the game.
