After completing Chapter 3, a trouble quest from Jolene should be available at the Trouble Center.
In her Help Wanted! quest, she'll ask you to clean out the storage room. You need to complete this quest and get your reward in order to start Prince Mush's comeback.
Help Wanted! Trouble Center Walkthrough
Some time after completing the Help Wanted! quest, Jolene will send Mario an email. In this email, she mentions that Prince Mush is planning his comeback.
Receiving this email takes around 20 minutes of active gameplay and moving between different locations.
Some time after receiving Jolene's email, Mario will receive an email from Prince Mush. In this email, Prince Mush tells Mario that he has completed his training and wants to fight Mario as his grand debut back at the Glitz Pit.
Prince Mush's email will come after about 20 minutes of active gameplay or more after receiving Jolene's email and fulfilling all requirements.
After you receive Prince Mush's email, you can fight him in an Exhibition Match at the Glitz Pit. Make sure you're well-rested, stocked on items, and have all the badges you need equipped before taking him on.
Do note that you do not need to become the Glitz Pit's champ again to fight Prince Mush.

Information: All attacks are ineffective when below half HP. Recommended Attacks: ・ Superguard first, then Hammer or Jump move to damage. |
Prince Mush will change his stance during your battle against him. Each stance has different attack conditions, so be prepared to use alternative attack strategies.
Note that while in his defense stance, you can only damage him after successfully using Superguard, which is when he kneels and his defense drops to 0.
Using attacks that ignore defense, like Koops' Shell Slam move, Madame Flurrie's Lip Lock move, or Piercing Blow badge, are very effective against Prince Mush's high defense of 6.
Jump attacks do not ignore defense, so if Prince Mush is in jump stance, you can use Charge to increase your next attack.
If you don't have a lot of Defense-piercing moves, you can use the special move Power Lift to boost the Attack and Defense of Mario and his partner. This will allow Mario and his partners to do some damage with non-piercing moves and take less damage from Mush's strong attacks.
Status effects from items, moves, or stage hazards cannot be inflicted on Prince Mush unless he's vulnerable after a Superguard. Before starting the match, make sure your inventory is full of recovery items instead of items that affect the opponent.
Successfully using Superguard against Prince Mush's combo attacks will make him kneel on the ground and drops his defense to 0. When he is on his knees, use your best jump and hammer attacks to deal high amounts of damage.
When Prince Mush's HP drops below half, he will enter defense stance, and the only way to damage him is to use Superguard successfully.
After taking some damage, Prince Mush will start using charged combo attacks that deal around 100 damage. If you try to avoid his attack after he charges, such as by using Vivian's Veil, he will just charge again and wait for you to appear before launching his attack.
Since you can't avoid this attack, the only way to stop it is to use Superguard. If you don't successfully Superguard his first attack, you can Superguard any of his subsequent ones in order to stop his onslaught.
Prince Mush also has the ability to spawn clones of himself, which you can defeat easily by using spread attacks. Note that if you ignore the clones, they can reduce your party's HP very quickly.
Always carry a lot of Life Shrooms when having a battle against a powerful boss, such as Prince Mush, to revive your knocked out characters.
This move involves walking to the center of the stage and delivering a punch. He will charge for a bit when gets to the center of the stage, so be careful of the timing of your Superguard.
This move is where he jumps and then charges forward while delivering a kick. This attack can also target the character positioned at the back, so be sure to watch the trajectory and determine the target of the attack.
This move is used after he uses charge for a moment, and is faster than a normal flying kick and is characterized by getting closer to the target. It is very powerful, and if you fail to Superguard, you can get knocked out.
Even if you fail to super guard the first hit, you can still super guard in the middle of a combo, so don't give up.
After defeating Prince Mush, talk to him in the arena corridor to get an important item, Prince Mush's Belt. This is a key item and has no in-battle effect.
