Each chapter in Paper Mario: The Thousand-Year Door has a major boss that Mario and his team need to defeat. These bosses have relatively high HP, have several attacks they can use, and some fights even have a gimmick to mix things up.
Generally, you should use your most powerful attacks to defeat these bosses when they are vulnerable.
With the exception of a few chapters, Mario will get Crystal Stars after defeating major bosses. Each Star will give him a new special move to use and increase his Star Power meter, which is needed to use said moves.
