Life Mushroom! is a Trouble Center side quest from the Puni Elder that can be accepted at the Trouble Center after Chapter 2 in Paper Mario: The Thousand-Year Door (TTYD). Keep reading for a full walkthrough of the Life Mushroom! side quest, its location, rewards, and how to unlock it!

Once you have a Life Mushroom in your inventory, all you have to do is go to the Great Tree in Boggly Woods and give it to the Puni Elder.

You can obtain a Life Mushroom from Hooktail Castle for free. It is located on the same platform that Koopook stays on in the Try to Find Me! Trouble Center quest. However, take note that this item may have already been used if you picked it up early on.
A Life Shroom can also be obtained early from Westside Goods, the item shop in western Rogueport. It can be purchased for 50 coins here, but this area is only accessible after you get Zess T. a new contact lens.
If you're doing this quest later on, it can be easily purchased at a cheaper price from the Twilight Town item shop for 40 coins after completing Chapter 4. It can also be obtained through late-game drops and rewarded upon earning 150 shop points.
Life Shroom Effect and How to Get

Life Mushroom! from Puni Elder will appear at the Trouble Center once you complete Chapter 2. To activate this request, select it from the bulletin board at the Trouble Center.
The quest must first be accepted from the Trouble Center bulletin board before you can do any of the steps to complete it.
To get to the Trouble Center, you must first get through Gus, the guard blocking the east side of Rogueport. You can either pay him coins every time you need to pass or defeat him in combat. Once Koops joins your party, defeating him becomes much easier.

The Puni Elder will reward you with 50 Coins once you complete the Life Mushroom! trouble side quest. This is enough to cover the cost of buying a Life Mushroom from the Westside Goods shop.
