Since Amleth has no way to increase her ATK aside from getting damaged, ATK runes would do her nicely, as well as CRIT runes. These should go along with PM. DEF Break and SPD runes.
For defensive runes, you will want to use CRIT RES, Debuff RES, P. DEF and M. DEF runes to help mitigate the damage Amleth takes and delay her [Immortality] last stand. Though, eventually, you might want to replace one of the DEF runes with EVD instead.

If you were unlucky enough at the start of your run to not get either Florence or Cordie, getting Amleth is another option especially since you can get two copies of her early on if you bought the First Achievement Bonus Pack.

Later on, Amleth can be a potent unit for early PvP since by that time most enemy teams focus on burst damage (Cordie and Florence), Amleth can withstand that easily with her invulnerability. Make sure to pair her with Minasumari though so you have a way of countering Nina who can steal Amleth's invulnerability buff.
Amleth Analysis and How to Play
Last Witch Standing
Amleth's passives give her the chance to not only survive a devastating attack that would wipe your entire party, but increase her ATK greatly enough that she could still win the match.
'Fulfiller of Oaths' makes sure she'll survive with just 1 HP, and leveling it up further will make sure she lives for at least a couple more turns.
Meanwhile, 'Ray of Light' increases her ATK up to 80% if you have it at a high level. So when she finally uses her Active Skills, there's a good chance you'll win the battle even if she's the only witch you have left.
Won't Start a Fight, But She'll Definitely Finish It
Though both her Active Skills look nothing special, especially in comparison to more popular DPS options like Florence or Cordie, these will shine once the battle gets tough and her passives are activated.
