The Second Prince of Flowing Sands and the King of Flowing Sands are located in the Sandgate Village area, pass the enemies near the Village Entrance Keeper's Shrine in Chapter 2.
To reach his boss arena, start at the Shrine, cross the bridge and turn right. Turn left upon reaching the wooden platform and pass through the village outskirts. The boss arena will be pass the gate to the right of the area with the two-headed enemies.

In your first encounter, the King of Flowing Sands stands atop pillars, throwing rocks at you. To bring him down, lure the Second Prince into attacking the pillars, breaking them and leaving the King vulnerable. Once on the ground, the King may occasionally stagger the Second Prince with his rock throws, giving you brief windows to heal or attack.
Once the King is vulnerable to attacks, prioritize killing him first. This will put the Second Prince in a rage, but he will drop the Second Rat Prince Spirit at the end of the fight.
You could just ignore the King and take down the Second Prince, but doing so will cause you to miss out on the Spirit drop, and you'll need to replay the game on New Game+ to get it.
All Missables and Points of No Return
The order in which you defeat the bosses in this fight influences the cutscene that plays before the battle with the First Prince of the Flowing Sands. While the cutscene will change, the fight itself and the rewards remain the same.
The Second Prince's combo that starts with the thrusting attack is his easiest move to punish. Keep your distance, run to your right to avoid the hits, then counter with a charged Heavy Attack or any other move of your choice.
The Second Prince has two delayed attacks you may underestimate, but it's essential to dodge these to get more opportunities to be on the offensive.
In one, he throws his mace and follows up with a long-range wave attack. In the other, he spins his mace and performs a jumping attack, followed by a delayed area-of-effect strike. Both can be dodged when you see them coming, allowing you to follow up with attacks.
