Rome specializes in having a strong Culture and Military with bonuses that feed off each other. However, they're better suited for a Military Legacy Path rather than Culture since they have ways to heavily boost their Legions and don't get Production Boosts for Wonders.

The first step when starting a new game is to choose your Leader and Civ combo. For Rome, you'll want to pick Leaders with bonuses that can buff up their Units and Cultural output.

A few examples of strong Leaders are Augustus, since he's Rome's signature Leader and plays extremely well around its Town focused empire, Lafayette, for extra Tradition Combat Strength buffs on Legions, and Trung Trac, who specializes in Commanders and can immediately settle with your first Legatus.

Use All of Rome's Traditions

Thanks to Rome's powerful Cultural output, you can quickly unlock Civics compared to most other Civs. Because of this, you'll want to prioritize unlocking all of Rome's Traditions so that you can increase the bonuses of the Forum and buff your Legions' Combat Strength.

Legions
Rome is one of the strongest military Civilizations due to their Legions gaining massive Combat Strength bonuses from Traditions. Aim to build multiple armies full of Legions to exploit their strength, and to also get Culture when you have the Cursus Honorum Tradition slotted in your Government.

Settle With Legatus Not Settlers

Legatus Settle
Instead of building Settlers, you should use a Legatus to settle a Town instead. They can do this every three levels so make sure they're in the thick of battle to get the experience they need to level up.

As a note, you should build at least one Settler and settle a Town nearby so that you can jump-start your empire.

Don't Convert Towns to Cities

How to Convert Towns to Cities
A bunch of Rome's bonuses benefit from having Towns instead of Cities. Unless you need the extra space for certain buildings or Wonders, you should aim to keep all Settlements outside of your Capital as Towns to maximize the bonuses that you can get.

Should You Convert a Town Into a City?

Always Build the Colosseum

Colosseum
Unlike most other Civilizations, Rome has a unique Narrative Event when they build the Colosseum, allowing them to gain one of two bonuses. Unless you desperately need the Happiness boost, you should always pick the "Staged Battle" so that your Legions can get a further +2 Combat Strength boost.
