Based on the Background Request Text Log, Isaac Clarke was born in the eastern seaboard region of the American Republic. His father, Poul Clarke, was an award-winning ship architect under the Galactic Union Merchant Marine Corps. Poul Clarke often took extended time off-world away from his family, meaning Isaac spent most of his childhood with his mother, Octavia Clarke.
Despite Poul's absence, Isaac followed in his father's footsteps by pursuing studies in electrical and mechanical engineering, with his high honors being enough to get him selected by a prestigious engineering school. However, due to his family's financial situation, which was in shambles due to Octavia Clarke selling off most of their estate to obtain a Unitology membership, he instead attended a lesser-known school with the help of scholarships and financial aid.
After graduating with high honors, Isaac decided to put his education to use by enlisting in the Merchant Marine. He was known for his resourcefulness and quickly rose through the ranks, eventually obtaining a high-profile position close to the major shipping lanes.
However, Isaac's career stagnated during this time due to his mother's deteriorating mental health, meaning he needed to take care of her and pay for her psychiatric treatments. While visiting his mother's psychiatric facility, Isaac would meet her to-be girlfriend, neuropsychiatrist Nicole Brennan. Isaac and Nicole would then live together for a while until Nicole moved out to serve on the USG Ishimura following a promotion.
In the Remake, Isaac's recollection in the Guest Consultant Suites in Chapter 10 reveals that he and Nicole fought prior to the outbreak on the Ishimura.
Isaac breaks the news to Nicole that his mother, Octavia Clarke, died, taking his father's life before her own. He starts blaming Nicole for the tragic event, as she mentioned that Octavia "was herself again" and "would be better at home."
Despite Nicole showing compassion and offering condolences, Isaac brushes her sentiments off and states that she "might as well have killed them [herself]." Not wanting to deal with him any longer, Nicole tells Isaac to go to hell before ending the call, leaving him alone to regret what he said.
In the game's opening cutscene, Isaac watches a concerning video message from Nicole, which tells him that the Ishimura is falling apart and something strange is going on inside. This message was likely one of Isaac's motivations for volunteering to join the USG Kellion's mission of investigating the communications blackout of the USG Ishimura.
As they reach the Ishimura and begin their docking procedures, the planet cracker's damaged guidance tethers would cause Isaac and his crew to crash-land into the ship, putting a rough start to their investigation of the Ishimura and Isaac's search for Nicole.
Chapter 1: New Arrivals Walkthrough
Isaac becomes separated from his crew inside the Ishimura's Flight Deck after an attack from multiple necromorphs. Only Isaac, computer specialist Kendra Daniels, and chief security officer Zach Hammond survived this initial incident, and they needed to find a way to escape the ship.
After Isaac performs a few ship repairs with the help of Kendra and Hammond, he heads to the Mining Deck to get an SOS Beacon and places it on one of the asteroids in the Mining Deck to call for help.
While the USM Valor managed to receive their distress signal, the trio soon found out that it also picked up and opened an escape pod containing their infected crewmate, Chen, causing Necromorphs to take over the military ship and making it crash into the Ishimura.
While obtaining a singularity core from the USM Valor to repair an escape shuttle off the Ishimura, Isaac would be contacted by Dr. Terrence Kyne, who believed he had an answer to stop the Necromorph threat for good, much to Kendra's skepticism.
After meeting up with Dr. Kyne, who believes that returning the Marker to Aegis VII would stop the necromorphs, Isaac helps the doctor with their plans by repairing the shuttle and helping them load the Marker into it.
However, right before Isaac boards the shuttle with Kyne, Kendra betrays the both of them by killing the doctor and hijacking the shuttle. She then reveals that her true mission was to fulfill an EarthGov operation to retrieve the Marker from the Ishimura.
Chapter 11: Alternate Solutions Walkthrough
After Kendra flies away with the Marker, Nicole suddenly contacts Isaac. She helps him recall the shuttle back to the Ishimura and reprograms it to fly toward the colony. Following Nicole's instructions, Isaac lands on Aegis VII and proceeds to bring the Marker back to its pedestal.
However, returning the Marker caused the planet's gravity tethers to malfunction, making its geo-orbital tectonic load begin flying toward Aegis VII. Before Isaac can fix the problem, Kendra reappears, locking Isaac in a room to hold Nicole hostage and steal back the Marker.
She then shows Isaac how delusional he is by pulling up the full version of Nicole's last video message, which reveals that Nicole had already died by the time he reached the Ishimura.
Chapter 12: Dead Space Walkthrough
Despite the groundbreaking revelations, Isaac knew he still had to escape Aegis VII, heading back toward the shuttle to hopefully reach it before Kendra. After approaching the landing pad, the Hive Mind revealed itself, killing Kendra and preventing Isaac from escaping.
After a long battle against the Hive Mind and its numerous Necromorph minions, Isaac triumphs and boards the shuttle, escaping Aegis VII. However, even after reaching a quiet region in space, Isaac still finds himself haunted by hallucinations of the dead Nicole.
