Slaying the Hero Valentin Marks the Point of No Return
Slaying the Hero, Valentin, marks the point of no return. Certain dungeons can still be accessed, letting you backtrack areas that you might have missed, but certain areas will be inaccessible in the current playthrough, preventing you from completing certain requests.

Play New Game+ To Access Missed Content
Should you wish to access previously missed content, you may do so by entering New Game+ and continuing your current playthrough. Notably, your equipment will carry over to the subsequent playthrough, though not all progress will be carried over.
How to Access New Game Plus (NG+)
Rescue Lou Quest
Rescue Lou can be treated as a separate game mode, in which you cannot head back to any part of the map and must traverse strictly on the given path on Luna Fraterna.
If you choose not to fight the optional bosses in Luna Fraterna during the Rescue Lou quest, you will miss out on the Tie That Binds boosters and other rewards.
Although the Tie That Binds Boosters do not carry over to your main save file, the Resurgence Core Blood Code does. This makes Rescue Lou worth doing if you plan on tackling New Game+ afterwards.

Lycoris' MissablesLycoris' Missables
You can miss Lycoris' second Blood Code if you complete the Slaying the Hero Holly quest before she has the chance to give it to you. Once this quest is finished, she will no longer be available as a partner.
It is highly recommended that you max out the proficiency of her Blood Codes and collect all the trade items she offers before completing Holly’s quest, as she becomes unavailable once the quest ends.

Noah's Missables
After completing the Slaying the Hero Valentin quest, Noah becomes inaccessible. This locks you out of his requests and their rewards, blood codes, as well as the trade items he offers through affinity progression.
His disappearance means you can no longer give him gifts to earn trading points. Make sure to complete all his requests, collect his blood codes, and give him the gifts he wants before starting the Slaying the Hero Valentin quest.
World Chests and Other Loot
Some formae and weapons are only available during their specific Eras, like the Zealot's Ceremonial Sword, which is made available during the quest Slaying the Hero: Holly Quest and Timeline Shift decision: Holly.
Since some loot cannot be found during the Free Exploration or Current era, it is imperative that you collect all the necessary items and gear in each hero's era before finishing their quests.

Anonymous 17 daysLycoris can't be given gifts, so no affinity for her. Her special weapon is obtained south of Debris Hill mistle after repairing Holly's timeline, on the pillar she appears the first time. Also Lycoris has only 2 Blood codes, C & B. As she can disappear after a certain point, get max proficiency with the first blood code as soon as she's on your team, then rest at a mistle to talk with her and get the second. Maxed both as soon as I could to see if there were more, but no.
