How to Open the Chest in Hubble Castle: Water Tower
Walk in a Cross Pattern From the Statue
To unlock the chest in Hubble Castle: Water Tower - Level 2, stand next to the statue, and, from there, go through the wall from west to east, then north to south.
The chest contains a bottle of Glowing Holy Water in the Past, which can be exchanged later on for a Lightning Staff. Once you return to the Present, you can unlock the chest again using the same method to obtain a Mink Mantle.

Bring the Glowing Holy Water to Estard Island
After defeating Hybris, return to Estard Castle and give the Glowing Holy Water to the man on the first floor of the southwest tower to obtain a Lightning Staff. In addition, you can watch an optional cutscene of Father Martyn if you stop by the church.
Chest Contains the Lightning Staff If Opened Only After the Hubble Questline
Alternatively, you can obtain the Lightning Staff from the chest if you unlocked it after completing the events of Hubble (Past), as the weapon will replace the Glowing Holy Water inside. Doing this will lock you out of the aforementioned events in Estard, however.

Past: East of Rucker Castle
Hubble Castle is located northeast of Hubble Region. The castle is first reached in the story after exiting Rucker Castle and heading east.
Present: Northwest Corner of the Map
Hubble Region is located on the northwest corner of the map. To reach Hubble Castle in the Present, simply sail northwest from Estard Island or Zoom to nearby regions first before sailing the rest of the way.
