To level up your character's Unique Skill during their Career, your character needs to have a high amount of fans and a green friendship gauge with Director Akikawa in the Senior Year.
There are different requirements for characters whose initial Track aptitude is the highest. If a character has a higher Dirt aptitude than Turf, or has a Career full of Dirt goal races like Haru Urara, they would need less fans to level up their unique skill.
When you meet the conditions on the day of the event, your character's unique skill will be leveled up.

It is important to note, however, that increasing your friendship with the Director (in this case, Riko Kashimoto) is not a requirement to leveling up your Unique Skill when running the Unity Cup scenario. As long as you reach the required Fans by Early April of Senior Year, you will level up your Unique Skill.

When a character becomes 3★, their base unique skill will be upgraded with a better version of it. From there, each star unlock will increase its level by 1. This upgraded Unique Skill can levele up further by hitting the Career goals mentioned in the previous section.
You can get Star Pieces for a character by getting them again by Scouting or exchanging Goddess Statues. You can get the Star Pieces for certain characters with other currencies like Friend Points and Club Points or by obtaining them as Event Point Rewards and the Prize Derby.

Trainees can also get the Unique Skills of their Legacy Umamusume, if they have the Spark for it. If the Legacy was a 3★ during their Career, they can pass down their upgraded Unique Skill.
However, unlike the Trainee's Unique Skill, they cannot be leveled up during Career mode.

All skills have a condition that must be met for your character to use them. Usually, they are related to their Style aptitudes.
Always check how your character's Unique Skill is activated in a race so you can pick skills to support or build it.

Wit is the stat that determmins how often your character will activate their skills. A 300 or C in Wit can almost guarantee a skill activation.
