How's Bowser's Health? is a special quest that must be completed before a certain point in the story. Once this quest is unlocked, it is highly recommended to do it immediately before progressing the main story any further.
Limited-time quests will disappear upon progressing the story, and you will be unable to complete them for the rest of your playthrough.
After you complete Jellyfish Island, you will be able to find this side quest on Wayaway Island. Enter Bowser's Castle in Wayaway Island in order to start How's Bowser's Health?.
If this side quest doesn’t appear for you after you return to Shipshape Island from Jellyfish Island, you may need to progress the story a little bit.
Travel to Wayaway Island, where you’ll see Starlow heading toward Bowser’s Castle. Follow him to the entrance and talk to him there. When he asks if you’d like to visit Bowser, select Yes.
Once inside Bowser’s Castle, use the nearby Green Pipe to fast-travel to Bowser’s Room (also known as Bowser's Castle/2F).
Talk to Starlow again at the door to Bowser's Room to trigger a lengthy cutscene. The quest will be completed after the cutscene.

After completing How's Bowser's Health?, Starlow witll reward you with Bowser Spikes. It is a Glove that occupies the Glove slot.
