Reaching the Game Over screen multiple times is the only way to change the difficulty in Mario & Luigi: Brothership. Learn how to change the difficulty and unlock other difficulty settings in this guide!
The only way to change the difficulty is through a Game Over screen. The other two difficulties won't be available at first, and will only appear after losing a battle multiple times.
Easy Mode and Cakewalk are the only other difficulties in the game. Standard, the default difficulty, is the hardest in the game and can't be made more challenging aside from giving yourself handicaps.
If you did pick one of the easier difficulties, it will revert to Standard after the battle. Easy Mode and Cakewalk make Mario and Luigi stronger for the current battle only.
Getting a Game Over multiple times in a single battle unlocks the Easy Mode difficulty. This makes Mario and Luigi stronger in your next attempt, but you can still choose to remain on Standard difficulty if you wish.
Losing a few more times after unlocking Easy Mode unlocks an even easier mode called Cakewalk, which tunes the fight into a piece of cake (walk?).
The actual hack to making battles easier is to get stronger yourself! Take time farming EXP and leveling Mario and Luigi up, farm coins and update your gear pieces to the strongest available, and master your opponent's moves.

If you want hard mode, equip the items that make you die in one shot or damage yourself. There you go. XD I forgot I had that on Mario and it made the third arc of the story WAY harder.
