After you complete Conductor Island, you will be able to find this side quest on Conductor Island. Talk to The Great Conductor near the lighthouse in order to start The Lonely Great Conductor.
If this side quest doesn’t appear for you after you return to Shipshape Island from Conductor Island, you may need to progress the story a little bit.

In order to start this quest, talk to the Great Conductor near the lighthouse of Conductor Island. Use the green warp pipe and head north to quickly get to this area.
Conductor Island Location Guide
The Great Conductor will ask for something sweet and cold to soothe his "headache" and grumbling tummy. Get some Potato Gelato at Heatfreeze Island to satisfy his craving.
Use the Return Pipe to go to Heatfreeze Island, then follow the ice cream signs west to reach the Potato Gelato shop at the top of the cliff.

Return to Conductor Island, then take the green pipe back up to the Great Conductor. Talk to him to give him the Potato Gelato and complete the quest.
