You can pick up any Mario & Luigi game and play them in no particular order, depending on what piques your interest. All games within the series feature the same Action-RPG elements with a few key differences in core gameplay mechanics, and a variety of humorous stories.

Combat in Mario & Luigi games have always been turn-based with incorporated QTE (Quick Time Event) actions to perform Jumps, Dodges, and Counter-Attacks. Brothership builds on this combat system with the addition of Battle Plugs, which provide supplementary attack damage to Mario and Luigi during their battles.
Continuing with the series' wacky adventures, Mario and Luigi are transported to Concordia, a world that was home to one mega-continent that has mysteriously fractured. Equipped with hammers and a stronger-than-ever sense of brotherhood, the two embark on Shipshape Island, aiming to reunite all the scattered islands and restore Concordia to its former glory!
Brothership's core gameplay revolves around exploring the vast ocean, discovering islands, and connecting them all to Shipshape Island. Each island comes with different challenges that the brothers have to overcome in order to form an Island Link and rebuild Concordia.
Every island you explore will be home to a variety of friendly faces, but the series' main supporting characters are Connie the socket-and-plug-themed Wattanist, and Snoutlet, a quirky character who insists they're absolutely not a pig!
Looking to foil the brothers' plans and prevent you from relinking Concordia is the scheming Zokket, whose electric presence is enough to leave you in a shock! But another sinister, spiky-shelled menace looms on the horizon…
