The first step to unlocking the Sealed Door is by purchasing Fireworks in Moleville. These can be bought from a mole for 500 coins. Once bought, proceed to the next step.
The purchased Fireworks then must be exchanged with a girl mole next to the house where you bought the Fireworks.
Once you have acquired the Shiny Stone, proceed to Monstro Town, where it must be used to enter a sealed door leading to Culex.
In Monstro Town, proceed to the sealed door that will react to the Shiny Stone. When interacted with, the door will be opened.
Proceeding inside will lead to a dimension with Culex in view. The boss will ask you if you wish to take up a challenge of combat against it.
The Shiny Stone will not be lost after opening the Sealed Door, so you can choose to sell it again to the Mole in Moleville to get some Carbo Cookies.
The Sealed Door in Monstro Town houses Culex, a secret boss that is best fought at party level 20 or above.
Culex is a difficult boss, so be prepared! If you are not ready yet, you can choose to decline the challenge and return at any time. Consider looking for the best equipment and forming the best party before fighting him.
Defeating him will reward you the Quartz Charm, one of the best accessories in the game.
