Unlocking Culex involves a side quest where you need to purchase Fireworks on Moleville for 500 Coins.
After purchasing the item, head inside the house next to where you bought the Fireworks. Trade it with the mole inside the house to get the Shiny Stone.
Head to Monstro Town and use the Shiny Stone on the sealed door. Entering the door leads to a different dimension with Culex in view.
Culex Secret Boss: How to Unlock and Beat
This side quest requires you to defeat Jagger, a former member of Bowser's Koopa Troop, in a boss fight at Monstro Town. Afterward, you will need to face Jinx in three succeeding boss fights that increases in difficulty.
Defeating Jinx rewards you with the Jinx Belt, an armor that prevents instant death.
In Rose Town, there is a gardener Toad looking for a Seed and Fertilizer.
The Seed is dropped by the Beezo that runs away after Megasmilax is defeated. The Fertilizer can be obtained after defeating Valentina, given by the Beezo that can be found by walking on air at the rightmost edge of the town of Nimbus Land.
The Lazy Shell (both weapon and armor variation) can be found in chests upon climbing the vine that grows after giving the items to the Toad.
How to Get the Lazy Shell Armor and Weapon
This side quest will activate once you've slept on a certain house in Monstro Town. As you are sleeping, the 3 Musty Fears will visit Mario and invite him to play their 'Find the Flag' game, starting the quest.
After you collect the flags, go back to Monstro Town and sleep again in the same bed where you started the quest. The 3 Musty Fears will then reward you with the Ghost Medal.
3 Musty Fears Flag Locations and Rewards
You can access the casino by obtaining the Bright Card from Knife Guy's Minigame. Afterwards, access the secret passage(one of the pipes) in Bean Valley by defeating a Piranha Plant.
Then inside, you will encounter a Chomp Chomp at the end. Defeat it and then at the corner located to the left of the Chomp Chomp, jump three times to reveal an invisible platform to get to the casino.
You can play a variety of minigames inside the casino but the one that you should look out for is Win Look the Other Way. To play the game, Mario simply has to look in the opposite direction of where Grate Guy will raise his finger.
Winning the Look the Other Way minigame will net random rewards. The most valuable item that can be obtained from playing Look the Other Way 100 times is the Star Egg.

The Cricket Jam is in a chest in Kero Sewers, in an area that can only be reached via Warp Pipe. The other end of the pipe is in a cave by the Sky Bridge in Land's End.
Give the Cricket Jam to Frog Sage to receive 10 Frog Coins!
Where to Get Frog Sage's Snack
The player will need to have returned to the Mushroom Kingdom from Marrymore after saving Peach from Booster to have access to this minigame.
After that, speak to the Beetle Mania Toad in the Inn located in Mushroom Kingdom and the minigame will be offered for purchase for 500 coins.
The goal in this game is to get as much points as possible. It is played by using the corresponding controls to shoot moving red Koopa shells and evade them! Unfortunately, there are no rewards for this game.
Beetle Mania: How to Unlock and Rewards
In Booster Pass, you'll find the Apprentices, Snifsters that wish to serve Booster.
To access their location, you'll have to go into Booster Tower and press the green switch there (you can't miss it when going up the tower). Back in Booster Pass, you'll then find the entrance to the training area where the Apprentices are.
The requirement to getting the Apprentices promoted is to lose to them, which is a tall order since they fight alone and have very weak stats. Have your party's HP get lowered elsewhere, then battle the Apprentice and make your whole party lose to it. The Apprentice will happily claim that it's now Snifster No. 4. You'll find it now in Booster Tower, just a couple of rooms before where you find Booster.
You can help five Apprentices achieve this (the succeeding ones will have higher numbers, e.g., Snifster No. 5). Leave the training area then return to find the next Apprentice. After helping the fifth and final apprentice (Snifster No. 8), you won't see anyone in the training area anymore, they're all in Booster Tower.
Aside from some chests in the training area, there are no rewards to this quest except for making these guys happy.
