The main way to get mules in your settlement is to order them via Pack Stations for 25 Regional Wealth once a month. Additionally, you can also order more frequently from Livestock Trading Posts.
When placing an order, remember to have vacant stable spaces provided by Hitching Posts to prevent Mules and other livestock from running away.

Sets up a Barter Connection, allowing you to send and receive goods between the current and another owned region. Employ Mules to transport up to 20 goods per trip. |
In Manor Lords, Mules are responsible for transporting items between settlements across two owned regions. This enables the Barter Connection that allows 20 goods to be transported between regions.
To be able to use Mules, players must also have Pack Stations set up between the two regions. The two Pack Stations will set up connections and act as the points of trade between settlements.
Mules are extremely helpful if you have set up multiple settlements in different regions.

Although Horses and Mules both provide logistical help, Mules are used to barter and transfer goods between the regions you own. Mules are also hired in Pack Stations and are working within owned settlements.
On the other hand, horses are employed to transport goods for trade to other regions. The horses are hired in Trading Posts where they make transporting goods for trade faster and more efficient.
