Select Casual Difficulty to have a faster time clearing the game. While it is possible to do so on higher difficulties, there are no actual achievements or trophies you can benefit from for speedrunning them.
You can still do it in other game difficulties if you want to challenge yourself and set a record.
Difficulty Setting Differences and How to Change

Similar to other RE titles, watching cutscenes won't stop the clock from ticking. Skip all the cutscenes and shave off a lot of time in your playthrough.

Some objectives and puzzle will take a lot of time to solve if you aren't familiar with them. Be sure to check the walkthrough and memorize them to progress quickly and save up some time.

The weapons in the Special Content menu are all powerful weapons. It is possible to speedrun without them, but it will take you less time to clear objectives that require killing, like fighting bosses and elite enemies.
You can try to complete challenges first to earn CP for unlocking them. You can do that by doing another run before you start speedrunning the game. You can also get in the Infinite Gun Ammo, so you don't have to spend time crafting or scavenging for resources.
Sneaking around is safer but it takes a lot of time just to pass through and get to the objective. If there are enemies that can't kill you instantly, it's better to run straight at them and take the damage if it can't be helped, rather than to crouch and waste time.

Opening and reading the files doesn't pause the game and will continue to run the timer. It's best to ignore them and focus on the objectives to clear the playthrough faster.

You can check the actual playthrough timer through the Challenges Menu. Scroll down until you see Speed Demon, where you can hover into to see the current time you have in your playthrough.
You can pause the game to stop the timer to take a break or check out the walkthrough for each section of the story. You can also look over the puzzles to solve them quickly.

You can get the Kotetsu for Grace to have an indestructible knife, which she can use to stop a grab attack against regular zombies. It can make a difference if you get caught by a zombie as it will shorten the animation.

You can get the Speed Demon Trophy is you finish the game within 4 hours. You will also get 20,000 CP, which you can spend on the bonus contents of the game.

You can also unlock the Infinite Ink Ribbon, which will allow you to save the game as Grace without a limit in Standard Classic or Insanity difficulty.

Finishing the game through speedrunning is required if you're aiming for 100% completion of the game. You will need to finish the game under 4 hours to get one of the trophies needed for 100% completion, which is the Speed Demon Trophy.
You don't have to do this if you just want to casually play the game and see the ending.
