Grace Ashcroft is one of the main protagonists in Resident Evil Requiem. She is a technical analyst for the FBI who is tasked on investigating the deaths in Wrenwood Hotel.

As she isn't used to combat, she has a different gameplay style in comparison to the Leon. Her gameplay portions play more like a classic survival-horror game where you need to sneak around to avoid enemy detection, solving puzzles, and being more vulnerable from threats due to lack of resources.
Her gameplay style also uses first-person view as default but you can switch to third-person.

Leon S. Kennedy is another main protagonist, who is also one of the main protagonists of previous games in the series (Resident Evil 2, 4, and 6). Leon is a federal agent of the D.S.O, a counterterrorism agency under the President of United States.
He was a former rookie police officer for Raccoon City Police Department and survived the Raccoon City incident back in 1998. After 30 years, Leon is dispatched to investigate the Wrenwood Hotel due to a case of a missing police officer. His story will be about him uncovering the truth behind the Raccoon City incident that has haunted his past.

As a hardened veteran, Leon's gameplay is more action oriented. Leon wields multiple firearms to fend himself from the infected, as well as equipped with a hatchet. Not only that, but he can also fight using melee attacks, parries, and even enemy weapons. His gameplay is set to third-person view as default but players can change it anytime.

Alyssa Ashcroft is the mother of Grace, and is a survivor of the Raccoon City incident. She was a reporter during the original outbreak and is one of the playable characters in Resident Evil Outbreak and File #2. She is a strong-willed reporter who is determined to investigate the truth about Umbrella until she was murdered in Wrenwood Hotel.
Victor Gideon is a researcher who kidnapped Grace, as he needs her blood to free Elpis. He is speculated to be the main antagonist of Resident Evil Requiem.
Nathan Dempsy is a FBI Special Agent who tasked Grace to investigate the murders in Wrenwood Hotel, which is tied to the events of Raccoon City in 1998.
