To beat Okita Soji, it's recommended to fight him as a solo boss during the Solitary Blade side quest. Follow these tips to easily win the encounter:

It’s highly recommended to take on Okita Soji during the Solitary Blade side quest because he appears as a solo boss in that encounter. If you defeat him there, he will not show up in the Crucible right before Tokugawa Yoshinobu.
Otherwise, you’ll be forced into a back-to-back fight against both of them. If you fall against Yoshinobu, you’ll have to defeat Okita all over again to get another shot.
You can safely jump over most of Okita Soji’s attacks because he lacks vertical reach. This is especially effective against his long attack chains and grabs. By jumping instead of blocking or dodging, you can conserve Ki to punish him from behind.
Keep an eye out for Okita Soji’s coughing fits, which typically occur after his long attack chains. Use this opening to safely deal damage, heal up, or refresh your buffs.
When attacking Okita Soji, be careful because he gains hyper armor during his own strikes, meaning he will not stagger. Make sure to disengage as soon as he starts his counter-attack or you’ll likely get punished.
Do not forget to use your summoning seals, especially as you can now have two of them equipped by this stage in the game. They can help deal extra damage, open opportunities to attack, or even save you from dying.
If you're struggling at the recommended level, take some time to gain a few extra levels and farm better gear before your next attempt.
How to Level Up Fast: Amrita Farming Guide
Keep your Living Artifact ready to secure the victory when Okita Soji is low. You're invulnerable while in this state but you must still dodge or block because sustaining damage shortens its duration.
You also have unlimited Ki while Living Artifact is active. Use this to spam your most potent Martial Arts.
Summon an acolyte from nearby benevolent graves to serve as a distraction and help deal chip damage. You'll need Ochoko Cups to summon acolytes, which you can obtain by defeating revenants from bloody graves.
Use talismans and items such as Power Talismans and Power Pills to boost your attack. Note that they do not stack and only refresh the duration of the attack buff.

Okita Soji is located in Mibu Temple and can be fought as a solo boss during the side quest Solitary Blade.
He will not show up later in the Crucible before the fight against Tokugawa Yoshinobu if you defeat him here.
If you didn't defeat Okita Soji in Mibu Temple, you'll encounter him in a back-to-back boss fight before Tokugawa Yoshinobu. This means you'll have to beat Okita every time you want another shot at Yoshinobu.
